How does skin and blood immunity affect development of melanoma

We are studying how the immune system both in the skin and in the blood can predispose people to developing melanoma.
We are studying a group of patients with Familial Melanoma Syndrome, who have a mutation in a vital gene which helps prevent cancer. This gene doesn't function as well for these patients and so they develop melanoma.
To see what differences in the immune system they have, we need to compare experiments done with their blood and skin to those of normal, healthy volunteers.
